108 Defining a bi-stable network switch that governs stem cell self-renewal and differentiation in squamous cell carcinoma

Stem cell – like tumor propagating cells self-renew to drive clonal expansion or differentiate into post-mitotic without tumorigenic potential in squamous carcinomas. This fate choice is governed by a transcriptional network comprised of SOX2-PITX1-TP63 driven self-renewal and KLF4 dependent differentiation circuits. Yet, how stem switch from remains elusive. Here, we report that this bi-stable Klf4 enhancer occupied the transcription factor SOX2 self-renewing differentiating carcinoma cells, on whether phosphorylated. We will present proteomic, transcriptomic, chromatin immunoprecipitation sequencing, conformation capture conditional gene targeting data leading discovery molecular mechanism governs self-renewal, differentiation, promotion growth. The identified provides novel path towards future development therapies could inhibit carcinogenesis patients.
